PROTOCOL FOR ISOLATION OF PHYTOPHTHORA (Erwin & Ribeiro, 1996).                     Download

  • Wash the diseased tissue thoroughly in running tap water.
  • Surface sterilize it with 0.1% mercuric chloride for 1min (time of exposure will change with the type of the tissue).
  • Wash it with three changes of sterile distilled water.
  • Blot it dry on a sterile filter paper.
  • Transfer it to selective medium for isolation of Phytophthora (PVPH medium).
  • Incubate it at room temperature for 5 days.
