DETECTION AND ISOLATION OF PHYTOPHTHORA FROM SOIL (ANANDARAJ & SARMA, 1990).

  • Sieve the test soil to < 2 mm fractions.
  • Place 25g of the sieved soil in a petri dish and add 40-50 ml of glass distilled water to make a soil-water suspension, so that 2-3 mm of free water remains above the soil when the particles get settled.
  • Float twenty leaflets of Albizia falcataria with the adaxial surface in contact with water.
  • Incubate the plates at 26o (± 2o C) for 72 h.
  • Surface sterilize the infected baits with 0.1% mercuric chloride for 1 min.
  • Rinse with three changes of sterile water and plate it on PVPH selective medium (Tsao & Guy, 1977).
  • This method can also be done in beakers instead of petri dishes.
