Procedure / Summary :

A modified DNA and RNA isolation method were standardized for the isolation of genomic DNA from P. colocasiae and RNA from taro. The genomic DNA isolation method involves the use of a cocktail of isolation buffer followed by very minimal isolation steps including centrifugation. The total RNA isolation involves the use of Trizol reagent coupled with chloroform. Later the mixture is subjected to centrifugation followed by precipitation with 100% ethanol solution.

Applications

The methods could be employed for all types of amplification of P. colocasiae genes and expression studies

Advantages

The entire process could be completed in less than an hour and yields DNA that is amenable for most downstream applications.The process is simple and amenable to be performed in a laboratory with minimal facilities.
