Detection of P. infestans load in soil using qRT-PCR protocols
Real time PCR quantification was carried out to determine the P. infestans load in the host tissues. Sampling was performed at different distance levels (5, 10, 15, 20 and 25 mm away from infected area) and different time intervals (24, 48, 72, 96 and 120 hrs of post inoculation). A maximum of 2.98 ng of P.infestans load was estimated at 5 mm away from the symptomic area while minimum 0.03 ng load was estimated 25 mm away from symptomic area in detached leaf experiment. This indicates that the P.infestans mycelium can penetrate the host tissue 25 mm away from the symptomic area. Similarly the quantification was estimated at different time intervals. Minimum pathogen load was 0.14 ng estimated at 24 hrs of intervals while maximum load was 2.29 ng estimated at 120 hrs of intervals.
The method can be used to detect oospores in the soil.
Robust and highly sensitive.
